The Bionova® BT30 Semi Self-Contained Biological Indicator (SCBI) is designed for accurate monitoring of dry heat sterilization processes at 160–180 °C. Each indicator contains Bacillus atrophaeus (ATCC® 9372) with 10⁶ spores per vial, providing a reliable challenge to sterilization cycles.

How It Works

  1. Place the BT30 indicator in the most challenging areas of the sterilizer load.

  2. Run the dry heat sterilization cycle at 160–180 °C as usual.

  3. After sterilization, carefully break the ampoule in tube B to allow contact between the spore carrier and culture medium in tube A.

  4. Incubate at 37 ± 2 °C for a maximum of 48 hours.

  5. Observe color change:

    • Blue → Yellow indicates sterilization failure (viable spores present)

    • Remains Blue confirms successful sterilization

  6. Positive control must turn yellow to validate results.

Directions & Safety Notes

  • Follow laminar flow or sterile environment procedures when handling ampoules and spore carriers.

  • Avoid exposure to temperatures above 180 °C or mechanical damage that could release spores externally.

  • Use the BT30 in combination with dual incubators (e.g., Bionova® IC10/20) for accurate temperature control.
